Target Information
Diacylglycerol (DAG) influences numerous cell signaling cascades by functioning as an intracellular activator of protein kinase C (PKC) and guanine nucleotide exchange factors. Intracellular DAG levels are regulated by diacylglycerol kinases (DGKs), which phosphorylate DAG to phosphatidic acid.
Human DGK isoforms:
- Type I DGKs: DGK-α (80 kDa), -β (90 kDa), -γ (90 kDa) — contain calcium-binding EF-hand motifs.
- Type II DGKs: DGK-δ (130 kDa), DGK-ι (130 kDa) — contain N-terminal pleckstrin homology (PH) domains.
- Type III DGKs: DGK-ε (64 kDa) — no identifiable regulatory domains.
- Type IV DGKs: DGK-Ω (104 kDa), DGK-ι (130 kDa) — possess C-terminal ankyrin repeats.
- Type V DGKs: DGK-θ (110 kDa) — contains 3 cysteine-rich domains and a PH domain.
