A yellow fluorescent protein (YFP) expression vector that eneables the co-expression of a gene of interest alongside the reporter. The plasmid also encodes the URA3 gene which is essential for uracil synthesis. This allows the plasmid to be maintained in cells in urcail deficient media. The cells must be deleted for the endogenous URA3 cassette to allow selection. The YFP reporter gene is called Kringle YFP and was developed by DNA2. 0. Reporter expression is controlled by the TPI promoter and transgene expression is controlled by the TEF1 promoter. These promoters demonstrate similar levels of expression.

Promoter Expression Level: This plasmid contains the yeast translation elongation factor 1 promoter. It is the strongest promoter that we provide for expression in Saccharomyces cerevisiae. It also contains the strong yeast constitutive triosephosphate isomerase (TPI1) gene promoter to drive the expression of the reporter gene. The TPI promoter demonstrates similar levels of expression to the translation elongation factor 1 promoter (TEF-1).
