This vector is designed for the expression of transgenes under the control of the CMV promoter for protein production in mammalian cells. The vector also contains a secreted luciferase reporter gene under the control of the RSV promoter. This allows for the detection of plasmid activity in the supernatant of mammalian cells.

Promoter Expression Level: This plasmid contains the mammalian CMV promoter to drive gene expression. We have tested all of our mammalian promoters in a range of cell types and CMV is consistently the strongest in those we have studied. However there are many reports of the CMV promoter demonstrating silencing by methylation in long-term culture. The RSV promoter driving reporter gene expression is one of the weakest mammalian promoters that we provide for many cell types but expression can vary significantly between cell types from different species. For example we have found that the RSV promoter shows significantly lower expression levels in human 293 cells when compared to hamster CHO cells.
