For IHC(P), use heat-induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen.
Target Information
BACE1 (beta-secretase) is a key enzyme involved in the production of amyloid beta-peptides (Abeta) found in extracellular amyloid plaques of Alzheimer’s disease (AD). Early-onset familial AD can be associated with a “Swedish” mutation in the amyloid precursor protein (APP), which enhances cleavage by BACE1.
Therapeutic strategies targeting BACE1 and other APP-cleaving enzymes such as gamma-secretase have been explored. APP is first cleaved by beta-secretase, producing a soluble derivative and a membrane-anchored C99 fragment. The C99 fragment serves as substrate for gamma-secretase to generate amyloid-beta peptide, deposited in the brains of Alzheimer’s patients.
BACE1 (also known as Asp2) is a transmembrane aspartic protease that colocalizes with APP.
