Description:
The LN3 monoclonal antibody reacts with human MHC class II molecule HLA-DR, expressed on antigen-presenting cells (APCs) such as B cells, monocytes, macrophages, dendritic cells, and activated T cells. HLA-DR is a heterodimeric transmembrane protein composed of alpha and beta subunits, playing a crucial role in peptide presentation to CD4⁺ T lymphocytes.
Applications Reported:
LN3 antibody is reported for use in flow cytometric analysis.
Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended usage is 5 µL (0.01 µg) per 100 µL test volume. Applicable cell number: 10⁵–10⁸ cells/test.
Fluorochrome Information:
Alexa Fluor™ 700 emits at 723 nm and is excited with a red laser (633 nm). Recommended optical setup: 685 LP mirror and 710/20 filter.

HLA-DR is a transmembrane glycoprotein composed of a 36 kDa alpha chain (DRA) and a 27 kDa beta chain (DRB).
- The alpha chain gene (DRA) has 5 exons: Exon 1 encodes the leader peptide, exons 2–3 encode extracellular domains, and exon 4 encodes the transmembrane and cytoplasmic regions.
- DRA lacks polymorphisms in the peptide-binding region and pairs with DRB1, DRB3, DRB4, and DRB5.
- DRB1 alleles determine peptide-binding specificity, crucial for transplantation typing.
- Expressed mainly on antigen-presenting cells such as B lymphocytes, monocytes, macrophages, thymic epithelial cells, and activated T lymphocytes.
- HLA-DR, DQ, and DP loci encode the main class II MHC molecules involved in antigen presentation to T-helper cells, initiating immune responses.
HLA and MHC antibodies are essential in Immunopeptidomics, aiding in neoantigen identification via LC-MS/MS.
