Target Information

Programmed death-ligand 2 (PD-L2), also known as B7-DC, is a member of the B7 ligand family within the immunoglobulin superfamily. Along with PD-L1, it acts as a ligand for PD-1. PD-L2 is mainly expressed in dendritic cells but can be induced in various immune and non-immune cells depending on the microenvironment. Expression increases with cytokines such as IL-4, TNF-alpha, and IFN-gamma. Although expressed at lower levels than PD-L1, PD-L2 has a 2–6 times higher affinity for PD-1. PD-1 and its ligands function as inhibitory immune checkpoint molecules, reducing T cell activation, proliferation, and cytokine secretion. PD-L2 may also stimulate T cell proliferation and cytokine production independently of PD-1, suggesting additional receptors. Recent studies show PD-L2 binds to RGMb, influencing respiratory tolerance via BMP and/or neogenin signaling. The human PD-L2 monomer consists of a 201-amino-acid extracellular domain, a 21-amino-acid transmembrane domain, and a 32-amino-acid cytoplasmic domain.
