Programmed death-ligand 2 (PD-L2), also known as B7-DC, is a member of the B7 ligand family within the immunoglobulin superfamily. Along with PD-L1, it acts as a ligand for PD-1, an inhibitory immune checkpoint receptor. PD-L2 is primarily expressed in dendritic cells but can be induced in various immune and non-immune cells depending on the microenvironment. Its expression is upregulated by cytokines such as IL-4, TNF-alpha, and IFN-gamma. Although generally lower in expression than PD-L1, PD-L2 binds PD-1 with higher affinity (2–6 times). PD-L2 interaction with PD-1 inhibits T cell activation and cytokine secretion, contributing to immune regulation. Additionally, PD-L2 can interact with RGMb, influencing respiratory tolerance and immune signaling.
