Product Specific Information

The PK136 monoclonal antibody reacts with mouse NK1.1, an antigen expressed by natural killer (NK) cells and a subset of T cells in NK1.1 mouse strains such as C57BL and NZB. Strains like BALB/c, SJL, AKR, CBA, C3H, and A do not express NK1.1 antigen.
For these strains, use monoclonal antibody DX5 (Product #14-5971) for NK cell detection.

Simultaneous staining of C57BL/6 spleen cells with PK136 and DX5 shows coexpression of both markers by most cells, with small populations of DX5⁺PK136⁻ and DX5⁻PK136⁺ cells.

  Optimal titration recommended for best assay performance.

Brilliant Violet™ 605 (BV605) is a tandem dye emitting at 605 nm, designed for cytometers with a 405 nm violet laser. Ensure instrument compatibility.

When using multiple Brilliant Violet™, Brilliant Ultra Violet™, or polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product #SB-4401-42) or Brilliant Stain Buffer™ (Product #00-4409-75) to minimize non-specific interactions.

Light Sensitivity: Protect vial and stained samples from light to avoid photo-induced oxidation.
Fixation: Samples can be stored up to 3 days in the dark at 4°C using IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54).
Compatibility: BV605 is not compatible with methanol-based fixation.

Target Information

NK1.1 (KLRB1, NKR-P1A, CD161) is a receptor expressed by Natural Killer (NK) cells, which mediate cytotoxicity and cytokine secretion.
The KLRB1 protein belongs to the C-type lectin superfamily, containing extracellular lectin motifs, a transmembrane domain, and a cytoplasmic domain.
It is classified as a type II membrane protein with an external C terminus.
The receptor recognizes Lectin Like Transcript-1 (LLT1) as its functional ligand.
