Product Specific Information

The PK136 monoclonal antibody reacts with mouse NK1.1, an antigen expressed by natural killer (NK) cells and a subset of T cells in NK1.1-positive mouse strains, including C57BL and NZB. Strains such as BALB/c, SJL, AKR, CBA, C3H, and A do not express NK1.1; for these, use the DX5 antibody (CD49b, clone DX5).

• Applications Tested: Tested on mouse splenocytes at ≤1.0 µg/test (100 µL final volume; 10⁵–10⁸ cells/test)
• Recommended: Titrate for optimal performance

Fluorochrome Information:
Brilliant Violet™ 605 (BV605) emits at 605 nm and is intended for use with a 405 nm violet laser. Ensure the cytometer is compatible with this fluorochrome.

Staining Buffer Recommendation:
When using polymer dye-conjugated antibodies (Super Bright, Brilliant Violet™, Brilliant UV™), use:

• Super Bright Complete Staining Buffer (Product # SB-4401-42), or
• Brilliant Stain Buffer™ (Product # 00-4409-75)
  to minimize non-specific interactions.

Light Sensitivity: Protect from light to avoid photo-induced oxidation.
Fixation: Compatible with IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark.
Not Compatible: Methanol-based fixation.

Target Information

NK1.1 (KLRB1, NKR-P1A, CD161) is a C-type lectin-like receptor expressed on NK cells and certain T cell subsets. It mediates cytotoxicity and cytokine secretion upon immune stimulation. The KLRB1 protein includes extracellular C-type lectin motifs, a transmembrane domain, and a cytoplasmic domain, classifying it as a type II membrane protein. Its receptor, NKR-P1A (CD161), recognizes Lectin Like Transcript-1 (LLT1) as its functional ligand.
