Target Information
Thioredoxins (Trx)는 산화환원 효소 활성을 가진 소형 다기능 단백질로, 모든 생명 세포에 존재합니다. Trx는 Cys-Gly-Pro-Cys 활성 부위 내의 redox-active disulfide/dithiol 그룹을 포함하며, 두 cysteine 잔기가 산화되어 이황화 결합을 형성할 수 있습니다.
Trx reductase는 NADPH를 전자 공여체로 사용하여 산화된 Trx의 활성 부위 이황화 결합을 환원시킵니다.
환원된 Trx는 DNA 합성에 필수적인 ribonucleotide reductase의 수소 공여체로 작용하며, 성장 및 redox 조절에 관여하는 일반적인 단백질 이황화 환원 효소입니다.
Trx의 주요 표적 단백질에는 protein disulfide-isomerase (PDI)와 p53, NF-κB, AP-1 등의 전사 인자가 포함됩니다. 또한 Trx는 methionine sulfoxide reductase 또는 peroxiredoxin isoform과 함께 작용할 때 H₂O₂ 제거 능력을 갖습니다.
