Product Specific Information
Description:
The H57-597 monoclonal antibody reacts with the beta chain of mouse TCR. TCR beta is expressed by thymocytes in a developmentally regulated manner and by most peripheral T cells. Crosslinking of the TCR complex with H57-597 induces activation, proliferation, or apoptosis depending on assay conditions. It serves as a phenotypic marker for TCR beta-expressing T cells.

Flow cytometric analysis.
Applications Tested:
Tested on mouse thymocytes and splenocytes at ≤ 0.6 µg per test (100 µL final volume, 10⁵–10⁸ cells/test). Titrate for optimal performance.

Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) enhances staining performance and should be added to antibody cocktails before mixing with cells (5 µL/sample).

Target Information
T cell receptors (TCRs) are essential for distinguishing foreign from self-peptides presented by MHC class II molecules, a key function in adaptive immunity. Dysregulated TCR recognition of self-peptides is linked to autoimmune diseases, while mutant self-peptides are associated with tumors. Superantigen-mediated TCR engagement can cause toxic shock syndrome. Autoantibodies to TCR V beta segments have been found in rheumatoid arthritis and SLE patients, potentially modulating TH1-mediated inflammatory responses.
Human T cells generally express TCR αβ with CD4 or CD8, while a smaller subset (double negative, DN) lacks both. γδ T cells, found mainly in epithelia, show limited TCR diversity and distinct antigen recognition, with subsets exhibiting antitumor and immunoregulatory functions.
