Product Specific Information

The H57-597 monoclonal antibody reacts with the beta chain of mouse TCR. TCR beta is expressed by thymocytes and most peripheral T cells. Crosslinking with H57-597 induces activation, proliferation, or apoptosis depending on assay conditions. It serves as a phenotypic marker for TCR beta-expressing T cells.

  • Applications Reported: Flow cytometric analysis
  • Applications Tested: Tested on mouse thymocytes and splenocytes, ≤0.4 µg/test
  • Recommended: Titrate carefully for optimal performance

Each product includes:

Compatibility Notice:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for NovaFluor conjugates.

CellBlox Plus Blocking Buffer Usage:
Add 5 µL per sample (10³–10⁸ cells) to antibody cocktails before combining with cells.
For single-color controls, use 5 µL per 100 µL of cell sample.

NovaFluor conjugates utilize Phiton technology based on nucleic acid dye structures, engineered for improved spectral performance and reduced spillover.

  • Excitation: 552 nm

  • Laser: 561 nm (Yellow)

Internal testing indicates strain-dependent non-specific staining of B cells in SJL mice; not observed in BALB/c or C57BL/6 strains.

Target Information

T cell receptors (TCR) are critical for distinguishing self from foreign peptides presented by MHC class II molecules, influencing adaptive immune responses.
Mutations and autoantibodies to TCR beta segments are linked to autoimmune diseases such as RA and SLE.
TCR diversity includes:

  • Alpha-beta T cells: Express CD4 or CD8
  • Double negative (DN) T cells: Lack CD4/CD8, increased in certain disorders
  • Gamma-delta T cells: Found in epithelia, show antitumor and immunoregulatory activity
