Target Information
MARCH8 (c-MIR) is a novel E3 ubiquitin ligase belonging to the modulator of immune recognition (MIR) family, with a variant RING domain (RING-CH domain).
It acts as a functional and structural homolog of KSHV MIR1 and MIR2, targeting B7-2 for lysosomal degradation and down-regulating its surface expression via ubiquitination.
MARCH8 also down-regulates transferrin receptor and Fas expression, contributing to apoptosis regulation.
It is the first known E3 ubiquitin ligase capable of inhibiting MHC II expression.
Recent studies suggest MARCH8 regulates immune responses by promoting ubiquitination and endocytosis of MHC-II and CD86.
