Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. Upon stress, p53 activates transcription of specific genes such as p21waf1, leading to G1 or G2/M arrest.

Structurally, p53 includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerization domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is crucial for activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation. p53 plays a key role in preventing proliferation of damaged cells and acts as an anticancer factor. It is also linked to aging, oxidative stress, and deficits in long-term potentiation related to learning and memory.

In normal cells, p53 is expressed at low levels, but is highly expressed in transformed cell lines, contributing to malignancy. Mutations in p53 often result in loss of DNA binding and tumor suppressor activity. TP53 gene alterations occur both as somatic mutations in cancers and as germline mutations in hereditary cancer syndromes such as Li-Fraumeni.
