Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation. p53 upregulation triggers transcription of genes such as p21waf1, leading to G1 or G2/M arrest. Structurally, p53 contains an N-terminal transactivation domain, a central DNA-binding domain, an oligomerization domain, and a C-terminal regulatory domain.
Phosphorylation at different sites, such as Ser15 and Ser392, plays key roles in p53 activation, stabilization, and tumor association. p53 acts as a critical tumor suppressor by preventing proliferation of damaged cells and has implications in aging, oxidative stress, and memory function. While normally expressed at low levels, it is upregulated in many transformed cells, contributing to malignancy. Mutations in the TP53 gene are common in human cancers and hereditary cancer syndromes like Li-Fraumeni syndrome.
