Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. Upon stress, p53 activates transcription of genes such as p21waf1, leading to G1 or G2/M arrest.

p53 structure includes:

• N-terminal transactivation domain
• Central DNA-binding domain
• Oligomerization domain
• C-terminal regulatory domain

Phosphorylation at specific sites, including Ser15 and Ser392, is crucial for p53 activation and stabilization. Ser392 phosphorylation has been associated with tumor formation. p53 also plays roles in aging, oxidative stress, and learning/memory processes.

In normal cells, p53 is expressed at low levels, but in transformed cell lines, it is highly expressed and may contribute to malignancy. Mutations in TP53 can lead to loss of DNA binding and tumor suppressor activity, contributing to various cancers and hereditary cancer syndromes such as Li-Fraumeni syndrome.
