Target Information
ZAP-70 is a cytosolic protein tyrosine kinase belonging to the Syk family. It is expressed in T-cells and natural killer cells and is essential for T-cell receptor activation.
Upon TCR engagement, ZAP70 is phosphorylated on Tyr292, Tyr315, and Tyr319 in the interdomain B region between the SH2 and kinase domains.
- Tyr315 (Vav binding site) and Tyr319 (Lck binding site) enhance ZAP70-mediated lymphocyte signaling.
- Tyr292 phosphorylation attenuates signaling by terminating transient activation.
