• Immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature (RT).
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at RT for 20 min.

Target Information

Alpha-2-Macroglobulin (A2M) is an approximately 720 kDa homotetrameric glycoprotein composed of four identical ~180 kDa subunits.
A2M shares structural similarities with other alpha-macroglobulins such as complement components C3 and C4 and pregnancy zone protein (PZP), and exhibits a broad ligand-binding capacity.
It functions as a humoral defense barrier against foreign peptides by capturing various proteinases, acting as a panprotease inhibitor.
In Alzheimer’s disease (AD) brains, A2M is localized in diffuse amyloid plaques, suggesting a role in AD pathogenesis.
