Product Specific Information
- Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 min.
Target Information
Alpha 2-Macroglobulin (A2M) is an approximately 720-kDa homotetrameric glycoprotein composed of four identical ~180 kDa subunits. A2M shares with other alpha-macroglobulins, such as complement components C3, C4, and pregnancy zone protein (PZP), an extraordinary binding capacity for various ligands. This enables alpha-macroglobulins to function as humoral defense barriers against foreign peptides in plasma. A2M interacts with and captures virtually any proteinase, acting as a panprotease inhibitor. In Alzheimer’s disease (AD) patients, A2M has been localized to diffuse amyloid plaques, suggesting a significant role in AD pathophysiology.
