Target Information
Arylalkylamine N-Acetyltransferase (AANAT) is the penultimate enzyme in melatonin synthesis. Hormonal melatonin is implicated in sleep and circadian clock function. Large changes in AANAT abundance are believed to drive the day/night rhythms in melatonin synthesis in the pineal gland and retina. Most research suggests AANAT activity is regulated by steady-state protein levels, but additional mechanisms may control melatonin production without altering AANAT protein or activity, potentially explaining the rapid increase in circulating melatonin at the onset of darkness in humans.
