Target Information
Glial cell line-derived neurotrophic factor (GDNF) plays key roles in the control of vertebrate neuron survival and differentiation.
A novel neurotrophic factor, neurturin (NTN), has been cloned from human and mouse. Physiological responses to neurturin require receptor tyrosine kinase RET and a glycosylphosphatidylinositol-linked receptor NTNRα.
The cDNAs encoding NTNRα from multiple species have been cloned and termed GDNFR-β, RET ligand 2 (RETL2), or TGF-β-related neurotrophic factor receptor 2 (TrnR2), and are now known as GFR-α2.
NTN binds to and forms a complex with GFR-α2 and RET PTK, activating the RET receptor tyrosine kinase pathway. Both NTN and GDNF can activate the MAP kinase and phosphatidylinositol 3-kinase pathways, playing critical roles in neuronal development.
Neurturin and GDNF define a new family of neurotrophic factors.
