Target Information

EIF2AK2 (PKR) is one of four kinases that specifically phosphorylate Ser51 of translation initiation factor eIF2-alpha in response to various environmental stresses.
PKR plays a key role in antiviral defense mediated through its activation by double-stranded RNA produced during viral infection. It is involved in IFN-induced innate antiviral response, virus-induced apoptosis, cell growth, and differentiation.
PKR induces apoptosis by up-regulating Fas expression and mediates FADD/Caspase-8 death signaling. Upon viral infection, PKR functions as a dual protein, sequentially activating both cell survival and cell death pathways through kinase-independent and -dependent mechanisms.
It regulates multiple pathways including NF-kappaB, p53, p38, and PDGF signaling, and has been implicated in tumor suppression and malignancy.
To evade PKR’s antiviral effects, viruses have evolved mechanisms such as inhibition by influenza virus NS1 protein. PKR is also associated with several neurodegenerative diseases including Alzheimer’s, Huntington’s, and amyotrophic lateral sclerosis.
