Target Information

Estrogen Receptor beta (ERβ, NR3A2) protein is approximately 55 kDa, though multiple isoforms ranging from 36–59 kDa have been described. Human ERβ was initially cloned and characterized from testis.
Estrogen receptors are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. These ligand-activated transcription factors bind estrogen, induce conformational changes, and dimerize to interact with estrogen response elements (ERE) in DNA.
ERβ can regulate gene transcription positively or negatively via coactivator or corepressor recruitment. Two different forms exist—ERα and ERβ—encoded by ESR1 and ESR2, respectively. ERβ binds estrogens with similar affinity to ERα and can inhibit ERα activity in reproductive tissues.
ERβ is widely expressed and acts as a potent tumor suppressor, implicated in prostate cancer, autism spectrum disorder, medullary thyroid carcinoma, De Quervain disease, and cardiovascular disease risk.
