Target Information
Estrogen Receptor beta (ERβ, NR3A2) is approximately 55 kDa, with multiple isoforms ranging from 36–59 kDa. Human ERβ was initially cloned from testis. Estrogen receptors (ER) are members of the steroid/thyroid hormone receptor superfamily and function as ligand-activated transcription factors. Upon estrogen binding, ER undergoes conformational change, dimerizes, and binds to estrogen response elements (EREs) in DNA, regulating gene transcription via coactivators or corepressors.
Two forms of estrogen receptor exist: alpha (ESR1) and beta (ESR2). ERβ binds estrogens with similar affinity to ERα, and some isoforms can inhibit ERα activity in reproductive tissues. ERβ is widely expressed across tissues and can act as a tumor suppressor, playing roles in diseases such as prostate cancer, autism spectrum disorder, medullary thyroid carcinoma, De Quervain disease, and cardiovascular disease.
