Product Specific Information
The C17.8 antibody reacts with the p40 subunit of IL-12 and IL-23, neutralizing their bioactivity.
For specific neutralization of IL-23 only, refer to clone G23-8 (anti-mouse IL-23 p19).
IL-12
- 70 kDa heterodimeric cytokine composed of p40 and p35 chains
- Secreted by activated monocytes, macrophages, and dendritic cells
IL-23
- Heterodimer composed of disulfide-linked p19 and p40 subunits
- Expressed by activated dendritic cells
Applications Reported: IP, Western blot, ELISA, neutralization, and intracellular flow cytometry.
Applications Tested: Verified by LAL assay for low endotoxin levels; tested for ELISA detection and bioassay neutralization of IL-12 p70 and IL-23.
At 62.0 ng/mL, the antibody inhibits by 50% the biological effects of 1 ng/mL mouse IL-12 p70 or IL-23 in bioassays.

Target Information
IL-12
Produced by dendritic cells, neutrophils, macrophages, and lymphoblastoid cells upon antigenic stimulation.
Encodes IL-12A (p35) and IL-12B (p40).
Promotes leukocyte migration to tumors and induces ischemic necrosis.
IL-23
Shares IL-12B subunit with IL-12 and includes IL-23A (p19).
Stimulates memory T-cells and supports Th17 cell functions, enhancing neutrophil chemokine and pro-inflammatory cytokine production.
