Target Information
The human UNG gene encodes both nuclear (UNG2) and mitochondrial (UNG1) forms of uracil-DNA glycosylase.
These forms are generated by alternative splicing and the use of two differentially regulated promoters (PA and PB).
UNG1 and UNG2 cDNAs are similar in size (2061 and 2058 bp, respectively), resulting in unresolved mRNAs in gel electrophoresis.
Nuclear UNG2 differs from mitochondrial UNG1 by 44 amino acids in the N-terminal sequence, which is not essential for catalytic activity.
UNG gene products play a major role in repairing mutagenic U:G mispairs caused by cytosine deamination, for example, removing misincorporated dUMP residues.
Expression of UNG1 and UNG2 varies by cell and tissue type and is regulated by the cell cycle.
UNG activity is generally higher in proliferating compared to nonproliferating cells and tissues.
