CD26 (dipeptidyl peptidase IV, DPP IV), also known as adenosine deaminase (ADA) binding protein, is a homodimeric atypical serine protease belonging to the prolyl oligopeptidase family.
CD26 is expressed on lymphocyte cells and is upregulated during T-cell activation. It is also found on activated B cells, natural killer cells, and epithelial tissues.
CD26 plays roles in T-cell activation, cell adhesion to extracellular matrix components (fibronectin, collagens), and HIV infection. It is identical to adenosine deaminase complexing protein-2 and the T-cell activation antigen CD26.
As an intrinsic membrane glycoprotein and serine exopeptidase, CD26 cleaves X-proline dipeptides from the N-terminus of polypeptides.
Alterations in CD26 peptidase activity are associated with malignant transformation, with enzymatic activity increasing with tumor grade and severity.
Expression changes have been observed in circulating monocytes and CD4+ T cells during rheumatoid arthritis and systemic lupus erythematosus.
