The Oncomine™ TCR Pan-Clonality Assay is a targeted next-generation sequencing (NGS) assay designed to detect T-cell diversity and measure clonal expansion or contraction with high accuracy. It also enables the detection of rare clones with exceptional sensitivity.

Primers are specifically designed to sequence the FR3-J regions of the TCR beta (TRB) and TCR gamma (TRG) chains. Using Ion AmpliSeq™ technology, sequencing of the CDR3 region of both chains occurs in a single reaction. When integrated with Ion Reporter™ Software, data analysis and report generation are streamlined and efficient.

Workflow and Compatibility

The kit provides reagents for library construction and dNTPs. Ion AmpliSeq libraries can be generated from DNA extracted from:

• FFPE tissue
• Fresh or frozen tissue
• Whole blood
• Peripheral blood leukocytes
• PBMCs
• Sorted T cells

Only 50 ng of nucleic acid is required. The assay is optimized for sequencing on the Ion GeneStudio™ S5 System using Ion 530, Ion 540, and Ion 550 chips, supporting up to 24 samples per run.

Why Reflex Test?

When primary testing does not yield desired results, a secondary assay targeting different regions of the V(D)J region may improve sensitivity. By sequencing the FR3-J regions of both TRB and TRG in a single reaction, this assay significantly reduces the need for secondary testing, particularly for hard-to-sequence samples.
