Product Specific Information

Description:
The HIR2 monoclonal antibody reacts with human glycophorin A, a sialoglycoprotein expressed by erythroid precursors and mature circulating red cells.

Applications Reported:
This HIR2 antibody has been reported for use in flow cytometric analysis.

Applications Tested:
This antibody has been pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (0.25 µg) per test. A test is defined as the amount of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically (10⁵–10⁸ cells/test).

Fluorochrome Characteristics:
Super Bright 780 is a tandem dye excited by the violet laser (405 nm) and emits at 780 nm. Recommended detection filter: 780/60 bandpass.

Usage Recommendations:

  • When using multiple Super Bright dye-conjugated antibodies, use Super Bright Staining Buffer (Product # SB-4401-42) to minimize non-specific interactions.
  • Compensation values may vary between beads and cells; it is recommended to set compensation using stained cells.
  • Protect the dye and samples from light due to photo-induced oxidation sensitivity.
  • Samples can be fixed with IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark.

Target Information

CD235a (Glycophorin A, GPA):
A single-pass membrane sialoglycoprotein expressed in mature erythrocytes and erythroid precursor cells. CD235a carries the M and N blood group antigens and provides a mucin-like barrier that reduces red cell aggregation. It also serves as a receptor for certain viruses and bacterial adhesins (e.g., Sandei virus, parvovirus, Streptococcus Hsa adhesin).
