Product Specific Information

Description:
The LN3 monoclonal antibody reacts with the human major histocompatibility complex (MHC) class II, HLA-DR. HLA-DR is expressed on human antigen-presenting cells (APCs) such as B cells, monocytes, macrophages, dendritic cells, and activated T cells. It plays an essential role in presenting peptides to CD4+ T lymphocytes.

Applications Reported:
This LN3 antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (0.125 µg) per test. A test is defined as the amount of antibody staining a cell sample (10⁵–10⁸ cells) in a final volume of 100 µL.

Fluorochrome Information:
Super Bright 780 is a tandem dye excited by a 405 nm violet laser and emits at 780 nm. Recommended filter: 780/60 bandpass. Ensure instrument compatibility.

Staining Recommendations:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Compensation Notes:
Higher compensation values may occur in the violet 450/50 channel using UltraComp eBeads ([Product # 01-2222-42]) compared to single-color stained cells. Set compensation with cells if necessary.

Light Sensitivity:
Protect vial and stained samples from light to avoid photo-induced oxidation.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark.

Target Information

HLA-DR is a transmembrane glycoprotein composed of a 36 kDa alpha chain (DRA) and a 27 kDa beta chain (DRB). The alpha chain gene contains 5 exons encoding leader peptide, extracellular domains, transmembrane domain, and cytoplasmic tail. DRA acts as the alpha chain for DRB1, DRB3, DRB4, and DRB5, while DRB1 polymorphisms determine peptide binding specificities.

HLA-DR is expressed mainly on antigen-presenting cells such as B lymphocytes, monocytes, macrophages, thymic epithelial cells, and activated T lymphocytes. DR, DQ, and DP loci encode major MHC class II products that bind processed peptides and present them to T-helper cells, initiating immune responses.

HLA and MHC antibodies are crucial in Immunopeptidomics for neoantigen identification via LC-MS/MS.
