Product Specific Information

The W6/32 monoclonal antibody reacts with human MHC class I antigens (HLA-A, B, C).
These antigens, associated with β2-microglobulin, are expressed by all human nucleated cells and play a key role in cell-mediated immune responses and tumor surveillance.
W6/32 recognizes a non-polymorphic epitope shared among HLA-A, B, and C loci and immunoprecipitates both 43 kDa and 11–12 kDa chains. Cross-reactivity is observed in baboon, rhesus, and cynomolgus monkey.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells.
Use 5 µL (0.25 µg) per test in a final volume of 100 µL. Suitable for 10⁵–10⁸ cells/test.

PerCP-eFluor™ 710 can replace PE-Cy5, PE-Cy5.5, or PerCP-Cy5.5.

• Recommended filters: 685 LP dichroic mirror and 710/40 band pass filter (695/40 acceptable alternative)

Fixation Stability:
Stable after staining when exposed to 2% formaldehyde overnight at 4°C. Evaluate for alternative fixation protocols.

Filtration: 0.2 µm post-manufacturing filtered.

Target Information

HLA-class I major histocompatibility (MHC) antigens are intrinsic membrane glycoproteins expressed on nucleated cells and associated with β2-microglobulin.
They present foreign peptides to cytotoxic T cells, playing a critical role in antiviral and antitumor immunity.
Human HLA-class I molecules include HLA-A, HLA-B, and HLA-C.
HLA and MHC antibodies are essential tools in immunopeptidomics for identifying and characterizing neoantigens via LC-MS/MS.
