Product Specific Information

The 12G5 monoclonal antibody reacts with human CD184 (CXCR4), also known as Fusin, LESTR, or HUMSTR. CD184 is a G-protein-coupled chemokine receptor with seven transmembrane domains, serving as a coreceptor for X4 HIV-1 entry into CD4+ cells. It is mainly expressed on naive T cells and is upregulated by PHA and IL-2 stimulation.

The 12G5 antibody partially inhibits chemotaxis and calcium influx induced by SDF-1, blocks CD4-independent HIV-2 infection, and inhibits CD4-dependent infection by certain T-tropic HIV-1 isolates.

Target Information

CXCR4 (Fusin, LESTR, HUMSTR) is a G-protein-coupled chemokine receptor with seven membrane-spanning domains, functioning as a co-receptor for HIV-1 and HIV-2 entry into CD4+ cells. It binds CXCL12/SDF-1 to trigger intracellular calcium increase and MAPK activation. CXCR4 also acts as a receptor for extracellular ubiquitin and bacterial LPS, playing roles in hematopoiesis, cardiac development, and inflammatory response.

Antibodies to CXCR4 can block HIV-1 and HIV-2 fusion and infection. The amino-terminal domain and second extracellular loop serve as HIV binding sites.
