Product Specific Information

The 188 monoclonal antibody was generated against and reacts with mouse B7-H4 (also known as B7S1, B7X). Cross-reactivity to other proteins has not been determined.
B7-H4 is a member of the B7 family reported to inhibit T cell activation, cell cycle progression, and IL-2 production. The ligand for B7-H4 has not been identified.
Simultaneous staining with clones 9 and 188 indicates distinct epitopes with no steric hindrance. Clone 188 stains mouse B7-H4-transfected cells but not spleen cells.
Applications reported include flow cytometric analysis, tested at ≤0.5 µg per test (100 µL final volume, 10⁵–10⁸ cells/test).
Excitation: 488–561 nm; Emission: 578 nm; Laser: Blue, Green, Yellow-Green.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

B7-H4 (B7x, VTCN1) is a costimulatory protein functioning as a negative regulator of T-cell-mediated immunity.
Though its receptor is unknown, B7-H4 delivers inhibitory signals preventing T-cell proliferation and IL-2 production.
Expressed on activated T-cells, B-cells, monocytes, and dendritic cells.
Aberrant expression is linked to lung, breast, and ovarian cancers.
Overexpression may suppress T-cell immunity and promote tumor progression.
VTCN1 is also a potential biomarker for early detection of ovarian cancer and renal cell carcinoma (RCC).
