Target Information
Nucleocytoplasmic transport is a signal- and energy-dependent process occurring through nuclear pore complexes in the nuclear envelope. Proteins containing a nuclear localization signal (NLS) require the NLS import receptor, composed of importin alpha and beta subunits (karyopherins). Importin alpha binds NLS-containing cargo in the cytoplasm, while importin beta docks the complex at the nuclear pore. In the presence of nucleoside triphosphates and Ran GTPase, the complex translocates into the nucleus, where importin subunits dissociate. Importin alpha delivers its cargo into the nucleoplasm, and importin beta remains at the pore, interacting with FG repeats of nucleoporins essential for translocation.
