Product Specific Information
- Recommended positive controls: Cell lines or frozen tissue (e.g., liver or colon)
- Expected to cross-react with multiple mammalian species due to sequence homology
- Store as a concentrated solution; centrifuge briefly before opening
- Recognizes conserved amino acid sequences (VQEALD, VQAALD) in MDR1 (~170 kDa) and MDR3 (~140 kDa) P-glycoproteins
- Boiling samples may reduce WB signal intensity
- For flow cytometry: Fix cells with 3.7% formaldehyde for 10 min at room temperature, then incubate with 5–10 µg antibody for 30–60 min at 4°C
Target Information
P-Glycoprotein is a member of the ATP-binding cassette (ABC) transporter superfamily. These proteins transport diverse molecules across cellular membranes and are grouped into seven subfamilies (ABC1, MDR/TAP, MRP, ALD, OABP, GCN20, White).
This protein belongs to the MDR/TAP subfamily, associated with multidrug resistance. It functions as an ATP-dependent efflux pump for xenobiotic compounds, reducing intracellular drug accumulation and mediating resistance to anticancer agents.
It also plays a role as a transporter in the blood-brain barrier.
