Target Information
TLR5 is a Type I membrane protein belonging to the Toll-like receptor family and participates in the innate immune response to microbial agents. It mediates detection of bacterial flagellins and acts via MyD88 and TRAF6, leading to NF-kappa-B activation, cytokine secretion, and inflammatory responses.
TLR5 is highly expressed in liver, detected in lung, and present at low levels in most other tissues. The TLR5 gene locus is associated with susceptibility to Salmonella. Mouse strains vary in resistance to S. typhimurium infection, classified as susceptible (BALB/c, C57BL/6, C3H/He), intermediate (DBA/2, C75L), or resistant (A, CBA). The MOLF/Ei strain shows high susceptibility, a unique TLR5 haplotype, and lower TLR5 expression.
A common TLR5 stop codon polymorphism that disrupts TLR5 signaling is linked to susceptibility to Legionnaires’ disease, underscoring its role in innate immunity.
