Target Information
Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, upon binding to estrogen, undergoes conformational change enabling dimerization and DNA binding to estrogen response elements (ERE). ERs regulate gene transcription through coactivator or corepressor recruitment.
Two forms exist: ER-alpha (ESR1) and ER-beta (ESR2). Alternative splicing produces at least four ER-alpha isoforms (66, 53, 47, 35 kDa).
Estrogen receptors are expressed widely across tissues, playing roles in sexual development, reproduction, bone metabolism, and are implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
