Target Information
Human Rad18 (hRad18) is a homologue of the S. pombe Rad18. In yeast, Rad6 and Rad18 play crucial roles in post-replication repair, filling gaps in daughter strands during DNA replication of damaged templates. The Rad6–Rad18 interaction is essential for lesion bypass, forming a tight complex through a conserved ring-finger motif. This complex binds damaged DNA sites via the single-stranded DNA binding property of Rad18, facilitating degradation of stalled replication components through Rad6’s ubiquitin-conjugating activity.
The human RAD18 gene maps to chromosome 3p24–25, a region often deleted in lung, breast, ovary, and testis cancers. It encodes a 495-amino-acid protein (~56 kDa) that is ubiquitously expressed in various tissues.
