Target Information
Apoptosis, or programmed cell death, is critical in development, tissue turnover, and tumor formation. The PDCD4 gene encodes a tumor suppressor protein whose expression increases during apoptosis and is often lost in breast, colon, lung, and prostate carcinomas. PDCD4 inhibits the helicase activity of eukaryotic translation initiation factor 4A and suppresses AP-1–mediated transactivation. Akt kinase regulates PDCD4 via phosphorylation, reducing its inhibitory effect on AP-1–responsive promoters. Two isoforms of PDCD4 are known.
