Description

This kit contains reagents to measure the activity of firefly luciferase in mammalian whole cell lysates. It includes a convenient lysis buffer, assay buffer, and firefly substrate, providing a complete system for intracellular detection of promoter or pathway activity.
Light output from the luciferase reaction is captured using a luminometer and correlates with the amount of firefly luciferase protein produced, allowing determination of promoter activity driving firefly expression.
The reagents are optimized for Thermo Scientific Firefly Luc Plasmids but can also detect other firefly or ATP-dependent luciferases utilizing D-luciferin as a substrate.

Scientific Background

Firefly luciferase is a 60 kDa protein naturally produced by beetles of the Lampyridae family (e.g., Photinus, Luciola).
Its bioluminescent signal originates from the oxidation of D-luciferin.
Light output measured by a luminometer correlates with luciferase protein levels and promoter activity.
