Target Information

Casein kinase II is a protein serine/threonine kinase present in all eukaryotic cells, suggesting fundamental cellular roles. The holoenzyme is a tetramer containing two alpha or alpha-prime subunits (or one of each) and two beta subunits. The alpha subunit serves as the catalytic unit, while the beta subunit’s function remains unknown.
CK2 functions as a tetrameric complex of two regulatory beta-subunits and two catalytic units (alpha and alpha’), forming homomeric or heteromeric structures. Although alpha and alpha’ subunits are catalytically identical, regulatory proteins such as cdc2 and Hsp90 preferentially bind to the alpha subunit.
CK2 phosphorylates several key intracellular signaling proteins involved in tumor suppression (p53, PTEN) and tumorigenesis (myc, jun, NF-kappaB). It also influences Wnt signaling via beta-catenin phosphorylation and the PI3K pathway via Akt phosphorylation.
