Product Specific Information
- Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes.
- After antigen retrieval, incubate samples with primary antibody for 10 minutes at room temperature.
- Suggested positive controls: colon, lung, or breast carcinoma tissues.
Target Information
COX2 converts arachidonate to prostaglandin H2 (PGH2), a key step in prostanoid synthesis, including inflammatory prostaglandins.
This process involves two reactions:
- Cyclooxygenase (COX) reaction converting arachidonate to prostaglandin G2 (PGG2)
- Peroxidase reaction reducing PGG2 to PGH2
COX2 is constitutively expressed in tissues such as endothelium, kidney, and brain, and up-regulated under pathological conditions like cancer and inflammation.
Up-regulation is associated with increased cell adhesion, phenotypic changes, resistance to apoptosis, and tumor angiogenesis.
In cancer cells, COX2 promotes prostaglandin E2 (PGE2) production, influencing motility, proliferation, and apoptosis resistance.
COX2 is naturally inhibited by calcitriol (active Vitamin D) and is a known target of NSAIDs such as aspirin.
