Description:
The R12-31 monoclonal antibody reacts with mouse RANK (Receptor activator of NF-kappaB), a member of the TNFR superfamily. RANK ligand (RANKL/TRANCE/OPGL) binds to RANK on dendritic cells, upregulating anti-apoptotic protein BcL-XL and supporting dendritic cell survival. The cytoplasmic domain of RANK interacts with TRAF2, TRAF5, and TRAF6. Overexpression of RANK activates NF-kappaB and JNK pathways. RANK–TRAF6 interaction activates NF-kappaB, while JNK activation is mediated via TRAF2 binding.

Applications Tested:
Tested by flow cytometric analysis of mouse RANK-transfected cells. Recommended use ≤1 µg/test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Titrate antibody for optimal performance.

Purity: Greater than 90%, as determined by SDS-PAGE.
Aggregation: Less than 10%, as determined by HPLC.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

RANK is a TNFR family member. The full-length mouse RANK cDNA encodes a type I transmembrane protein with extracellular and cytoplasmic domains, containing two potential N-linked glycosylation sites. The extracellular domain includes four cysteine-rich repeats, sharing 81% amino acid identity between human and murine RANK.
