Target Information
Dynamin is a GTPase required for endocytosis of clathrin-coated vesicles from the membrane. Its role in endocytosis was first identified in studies of the temperature-sensitive shibire mutation in Drosophila melanogaster.
Three closely related isoforms have been identified in mammals:
- Dynamin 1: expressed exclusively in neurons
- Dynamin 2: ubiquitously expressed
- Dynamin 3: expressed primarily in testes
Each isoform has several splice variants, suggesting distinct intracellular targets or vesicle functions.
Common domain features include:
- N-terminal GTPase domain
- Central pleckstrin homology domain
- Proline-rich C-terminal region with multiple SH3 domains
These SH3 domains target dynamin to clathrin-coated pits. Dynamin interacts with proteins such as the AP2 adaptor complex and amphiphysin.
