Product Specific Information
For reconstitution, add 100 µL distilled water to reach a final antibody concentration of about 1 mg/mL.
For conjugation experiments, perform an additional desalting step using Zeba Spin Desalting Columns (7K MWCO, 0.5 mL, Product #89882).
Target Information
The KRAB (Kruppel-associated box) domain is a transcriptional repression domain found in numerous transcription factors. Over 220 KRAB-zinc finger protein (KRAB-ZFP) genes have been identified in the human genome. These proteins repress transcription via interactions with KAP-1 (KRAB-associated protein 1), an 835 amino acid polypeptide containing a RING finger, B boxes, and a PHD finger.
KAP-1 forms complexes with KRAB-domain transcription factors to enhance repression efficiency and interacts with HP1 (heterochromatin protein 1) and KRAZ1. KAP-1 targets KRAZ1 to centromeric heterochromatin regions containing HP1alpha, regulating transcriptional repression. Mutant KAP-1 unable to bind HP1 causes random KRAZ1 distribution and strong transcriptional activation.
