Target Information

CDK4/cyclin D3 serine/threonine kinase is a dimeric complex involved in controlling cell proliferation during the G1 phase of the cell cycle.
CDK4 forms a complex with D-type cyclins and is inhibited by p16. It mediates phosphorylation of the C-terminal region of Rb protein, leading to transcriptional repression of the E2F complex.
CDC37 and HSP90 associate with CDK4 not bound to D-type cyclins.
SMAD3 is a major substrate of CDK4 and CDK2.
CDK6 and CDK4 phosphorylate Rb protein at different residues (CDK6: Thr821, CDK4: Thr826).
