Product Specific Information
Description:
The H57-597 monoclonal antibody reacts with the beta chain of mouse TCR. TCR beta is expressed by thymocytes and most peripheral T cells. Crosslinking of the TCR complex with H57-597 induces activation, proliferation, or apoptosis depending on assay conditions. It serves as a phenotypic marker for TCR beta-expressing T cells.

Reported for use in flow cytometric analysis.
Applications Tested:
Tested by flow cytometric analysis of mouse thymocytes and splenocytes. Recommended use ≤ 0.6 µg/test in 100 µL final volume. Cell number can range from 10⁵ to 10⁸ cells/test. Titrate antibody for optimal performance.
Each product contains:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Important Notes:
- NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
- Add 5 µL per sample or per 100 µL of cell sample (10³–10⁸ cells) for single-color controls.
- Add buffer to antibody cocktails before mixing with cells.

Target Information
T cell receptors (TCRs) discriminate foreign from self-peptides presented by MHC class II molecules, crucial for adaptive immunity. Self-peptide recognition is linked to autoimmune diseases, while mutated self-peptides are associated with tumors. Superantigen engagement can cause toxic shock syndrome. Autoantibodies to TCR V beta segments have been found in rheumatoid arthritis and systemic lupus erythematosus, potentially modulating immune responses.
Human T cells primarily express TCR alpha-beta with CD4 or CD8. Double-negative (DN) alpha-beta T cells are increased in certain autoimmune and immunodeficiency disorders. Gamma-delta T cells, found mainly in epithelia, exhibit limited TCR diversity and unique antigen recognition, contributing to antitumor and immunoregulatory functions.
