Target Information
Ubiquitin-like protein ISG15 plays a key role in the innate immune response to viral infection either via its conjugation to a target protein (ISGylation) or as a free protein. It is secreted from monocytes in response to type I interferons and promotes natural killer (NK) cell proliferation and non-MHC-restricted cytotoxicity. Its synthesis is stimulated by IFN-α, IFN-β, or IFN-ω, but not IFN-γ. ISG15 expression can also be induced by overexpression of interferon regulatory factors or influenza B virus.
ISG15 is secreted by monocyte, T-lymphocyte, B-lymphocyte, fibroblast, and epithelial cell lines. It enhances NK cell proliferation, augments cytotoxicity, and induces IFN-γ from T cells, suggesting its role in amplifying immunomodulatory effects of interferons. Intracellularly, ISG15 functions as a ubiquitin homolog. Mutations in the ISG15 gene can result in Immunodeficiency 38 with basal ganglia calcification (IMD38).
