Target Information
Tumor necrosis factor (TNF) receptor associated factors (TRAFs) are adaptor proteins linking the TNF receptor superfamily to signaling pathways, playing crucial roles in regulating cell death and stress responses. TRAF proteins share a conserved region enabling interactions with cell receptors and other TRAFs, activating distinct signaling cascades.
TRAF2 and TRAF3 bind directly to CD40, a receptor involved in B cell immunity and NF-κB activation in mouse B lymphocytes. TRAF2 and TRAF6 are essential for CD40 signaling in non-hematopoietic cells. TRAF2 also interacts with lymphotoxin-beta receptor (LTβR) alongside TRAF3 and apoptosis regulators cIAP1 and Smac.
