Product Specific Information
The eBio64DEC17 antibody reacts with human IL-17A and functions as a neutralizing antibody.
IL-17A is a CD4+ T cell-derived cytokine that promotes inflammatory responses and is elevated in conditions such as rheumatoid arthritis, asthma, multiple sclerosis, psoriasis, and transplant rejection.
Human IL-17A exists as glycosylated 20–30 kDa homodimers, produced by activated peripheral blood CD4+ T-cells. It enhances ICAM-1 expression in fibroblasts and stimulates epithelial, endothelial, or fibroblastic cells to secrete IL-6, IL-8, G-CSF, and PGE2.
IL-17A also supports proliferation and differentiation of CD34+ hematopoietic progenitors into neutrophils.
IL-23-dependent, IL-17A-producing CD4+ T cells (Th17 cells) represent a distinct subset from Th1 and Th2 cells. IFN-γ and IL-4 negatively regulate Th17 differentiation.
Intracellular staining by eBio64DEC17 identifies the same cell population as eBio64CAP17, as confirmed by co-staining experiments.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated for intracellular staining of stimulated human peripheral blood cells.
Recommended usage: 5 µL (0.125 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
Excitation: 488 nm Emission: 695 nm Laser: Blue Laser.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
Interleukin-17A (IL-17A, CTLA-8) is a 32 kDa disulfide-linked homodimer (136 amino acids) that signals through IL-17R/CDw217.
It regulates NF-κB and MAPK pathways, stimulates IL-6 and COX-2 (PTGS2) expression, and enhances nitric oxide production.
IL-17A plays a key role in inflammatory and autoimmune responses.
